Heurstic search for metabolic engineering: de novo synthesis of vanillin

چکیده
We present the application of heuristic search to in silico metabolic pathway engineering. In particular, we discuss a new computational approach to elucidate complex pathways and to address the practical challenge of combinatorial complexity in pathway inference. We have implemented this approach in a new computational framework, called PathMiner, which is useful for designing metabolic engineering strategies. In this paper, we describe our approach to analyze pathways for the de novo synthesis of vanillin, as well as a transgenic strategy t s a o ©

Improved vanillin production in baker's yeast through in silico design
BACKGROUND Vanillin is one of the most widely used flavouring agents, originally obtained from cured seed pods of the vanilla orchid Vanilla planifolia. Currently vanillin is mostly produced via chemical synthesis. A de novo synthetic pathway for heterologous vanillin production from glucose has recently been implemented in baker's yeast, Saccharamyces cerevisiae. In this study we aimed at engi...
متن کاملDeregulation of S-adenosylmethionine biosynthesis and regeneration improves methylation in the E. coli de novo vanillin biosynthesis pathway
BACKGROUND Vanillin is an industrially valuable molecule that can be produced from simple carbon sources in engineered microorganisms such as Saccharomyces cerevisiae and Escherichia coli. In E. coli, de novo production of vanillin was demonstrated previously as a proof of concept. In this study, a series of data-driven experiments were performed in order to better understand limitations associ...
متن کاملMimicking a natural pathway for de novo biosynthesis: natural vanillin production from accessible carbon sources
Plant secondary metabolites have been attracting people's attention for centuries, due to their potentials; however, their production is still difficult and costly. The rich diversity of microbes and microbial genome sequence data provide unprecedented gene resources that enable to develop efficient artificial pathways in microorganisms. Here, by mimicking a natural pathway of plants using micr...

The effects and interaction between cAMP-analogue dibutyryl-cAMP and calmodulin antagonists were investigated on de novo synthesis and secretion of lipids in cultures of hepatoma McArdle-RH7777 cells and normal rat hepatocytes. Dibutyryl cAMP caused a significant decrease in the secretion of de novo synthesized triacyl [3H] glycerol in both cultures of McArdle cells and rat hepatocytes. The ...
